**Advertised Summary Job Description**

The Senior Research Worker will be responsible for coordinating tissue culture services in the Diabetes Research laboratories of the Division. These services include maintenance and culture of human inducible Pluripotent Stem Cells, and their differentiation into specialized organ and cell types using a variety of protocols. Most importantly, the incumbent will be responsible for providing a steady supply of gut organ cultures to be used for a variety of experimental designs. In addition, the incumbent will work closely with post-doctoral scientists to develop, refine, and optimize methods to culture monolayers of flow-sorted specific cell types derived from gut progenitor and stem cells. These duties are expected to require 50% time effort and will include mentoring of junior members of the laboratory, visiting students, and collaborators. In addition, the incumbent will perform the necessary quality controls to monitor the material delivered to post-doctoral scientists. The remaining 50% time effort will be devoted to the generation, maintenance, and analysis of primary mouse hepatocytes for a variety of physiological measurements such as glucose and lipid production, lipoprotein secretion, protein synthesis. The candidate will be involved with DNA and protein blot analysis, RNA and DNA extraction and PCR analyses (genomic and RT-PCR), maintenance of cell stocks, DNA transfection and reporter gene assays, chromatin isolation, protein purification, RIAs and ELISA measurements of metabolites and hormones, simple recombinant DNA techniques required for vector construction, plasmid DNA isolation, generation and maintenance of adenoviral stocks. The incumbent is also expected to assist in data collection. S/he will be responsible for maintaining clear and updated records, coordinating laboratory orders/stocking of supplies, radioisotope inventory/safety surveys, monitor and enforce compliance with CU regulations on fire and hazardous materials, and be responsible for equipment maintenance. Performs additional related duties as assigned.

**Minimum Qualifications for Grade**

Requires a Master's Degree and at least three years of experience, or a bachelor's degree and at least five years of related experience or equivalent in education, training and experience.

The candidate selected for this position is required to undergo a drug screening test prior to a final offer of employment being made.
